Upon encountering an antigen-presenting cell (APC), T helper cells get activated. These APCs present antigens along with necessary co-stimulatory signals, setting in motion a cascade of events that translate into the body's defense mechanism. The response is two-pronged, involving a humoral response that deals with pathogens outside of cells, primarily through antibodies, and a cell-mediated response that targets infected cells or intracellular pathogens.

After activation by APCs, T helper cells undergo a process called differentiation. This crucial process is influenced by the cytokines present in the environment around the T helper cells at the time of activation. Cytokines are signaling proteins that mediate and regulate immunity, inflammation, and hematopoiesis.

Depending on the cytokines, T helper cells can transform into various effector T cell subsets.
