During transcription, the RNA polymerase encounters a specific sequence in the RNA that forms a hairpin loop structure followed by a sequence of uracil nucleotides. The hairpin loop causes the RNA polymerase to pause momentarily. The series of uracil nucleotides forms weak hydrogen bonds with adenine nucleotides on the DNA template. These weak bonds make the RNA strand more prone to detachment.
As a result, the RNA polymerase disassociates from the DNA template, releasing the newly synthesized RNA molecule. Intrinsic termination is an efficient way for cells to regulate gene expression by ensuring that transcription terminates at precise locations within the genome.
